PhD Position on Innovative Catalytic Strategies to Combat Cancer (P2106)

The Swiss Nanoscience Institute (SNI) at the University of Basel invites highly motivated scientists to apply for the SNI PhD program in Nanoscience.Your positionWithin this PhD project, we aim to attach metal-containing cofactors (Mcof) to the surface of cancer cells, and to use their catalytic properties to uncage specific drugs. With this ambitious goal in mind, the Mcof will be equipped with a high-affinity antagonist that binds tightly to proteins that are overexpressed on the surface of cancer cells.To evaluate the therapeutic potential of this strategy, we have selected somatostatin, a G-coupled protein receptor that is highly present on the surface of various forms of cancer. Cyclic disulfide-bridged peptides (Cyc-peptS–S), developed in the group of Prof. Fani, display exquisite affinity and selectivity for somatostatin receptors. Upon covalent linking of the peptide with the metal cofactor to afford Cyc-peptS–S–Mcof, we anticipate that the catalytically active cofactor will accumulate in the proximity of cancer tissues that display multiple copies of the somatostatin receptor. Upon addition of a caged drug, the catalytic activity of the cofactor Cyc-peptS–S–Mcof will lead to the site-specific uncaging of the drug where its action is required (an illustration can be found in the project description at www.phd.nanoscience.ch). In the past years, the Ward group has pioneered several catalytic strategies that lead to the uncaging of various fluorescent probes and drugs in the presence of cells. Reactions include olefin metathesis, transfer-hydrogenation, allylic substitution, hydroamination reactions, etc. In the course of his/her PhD, the researcher will work in a highly stimulating environment at the interface between chemistry, biology and medicine. You will use state of the art metabolomics and metagenomics technologies, and learn multiple techniques related to bone and microbiome physiology from clinical data to pre-clinical intervention in a model of osteoporosis. You will work in the Musculoskeletal Physiology and Nutrition group, a dynamic and enthusiastic team of PhD researchers striving to find nutritional solutions to preserve bone health during aging.  Your PhD project:You will investigate in postmenopausal women the association between bone clinical data, circulating microbial metabolites and feces bacterial species. Based on these results you will select in an in-house database a list of probiotic(s) able to impact bone remodeling.  Molecular and bone cellular mechanisms of these probiotics will be investigated in in-vitro models. In the project you will finally assess the efficacy of these probiotic(s) to limit bone loss with age in a model of estrogen deficiency. Through this work, the student will collaborate with a clinical service at UNIGE to link metabolomics and metagenomics with clinical outcomes. Eawag conducts research, education and expert consulting to achieve the dual goals of meeting direct human needs for water and maintaining the function and integrity of aquatic ecosystems.The Department of Urban Water Management (SWW) located in Dübendorf (Zurich) offers aPhD position in Environmental Engineeringin the project Heat-Down: Integrated Modelling of Stormwater and Urban Heat for cooling Cities. The overall goal of the project is to gain new insights into the potential of using stormwater to mitigate urban heat in cities! The PhD project will involve the following tasks: (i) Modelling the dynamics of stormwater management systems and practices; (ii) Assembling a “total urban water cycle model”; (iii) Support the full integration of stormwater and urban microclimate models, and (iv) model setup of case studies. To develop these tasks, the PhD student will work closely with a Postdoctoral researcher also funded by the project, who will mainly focus on the microclimate modelling aspects. In addition, the PhD student will count on the support of the other project partners, namely Dr Kerry Nice (University of Melbourne, Australia), Prof. Frans van de Ven (TU Delft, The Netherlands) and Dr Cintia Dotto (Melbourne city, Australia), emphasising the interdisciplinary character of the project.The position is fully funded by the Swiss National Science Foundation for a duration of four years.
